State police spokeswoman Darcy Wells said the accident occurred on Interstate 87, also known as the Adirondack Northway.
Police said in a statement the bus had one operator and 55 passengers, while tour officials said all aboard were from Quebec.
"There is one confirmed fatality," the statement said. "Several others are being treated for mostly minor injuries at three area hospitals."
The tour operator said the bus was travelling from Quebec City to New York City. Josiane Grimard of JaimonVoyage.com said the bus crashed at 7:30 a.m.
Company officials had no word on the cause of the accident.
In an interview with The Canadian Press, the tour operator's marketing director said it was a full bus with more than 50 passengers and a tour guide aboard the bus.
Nancy Trudel said the bus picked up travellers in Quebec City, Trois-Rivieres and Longueuil overnight before embarking for a weekend in New York City.
Trudel said the company's owner, Andre Poulin, was en route to the accident site to help the passengers and provide any support he can on site.
"For us, our priority is the passengers," Trudel said. "But we don't know what happened or how the accident occurred."
While the tour is operated by JaimonVoyage.com, Trudel said the charter bus is operated by Autobus Fleur de Lys, based near Quebec City.
A bus company official said it would have more to say later Friday.
A number of local rescue squads were sent to the scene, where the bus is on its side in the median.
Wells said the accident occurred between exits 29 and 30 in North Hudson, which is about 150 kilometres north of Albany.
